Pre-clinical test systems (in vitro)
All cell cultures use human cells or whole-blood from healthy volunteers.
- Activation of human neutrophil granulocytes (e.g. oxygen radical synthesis via chemiluminescence or elastase release)
- radical scavenging by the test substance (chemiluminescence and or colorimetry)
- Lymphocyte proliferation (MTT)
- Lymphocyte proliferation including the measurement of cytokines (e.g. GM-CSF, IFN , IL-2, IL-4, IL-5, IL-6, IL-10, chemokines, etc.)
- Mediator synthesis of leukocytes in whole blood cultures (various, as in D); ELISA)
- Prostaglandin / leukotriene synthesis in whole blood cultures (various; ELISA)
- Mediator synthesis in monocyte / macrophage cultures (various; ELISA) either using different human cell lines or whole blood
- Histamine release (basophils: isolated cells / whole blood; ELISA or RIA)
- Cytotoxicity in vitro (cell line, freshly isolated mononuclear cells and or whole-blood; trypan bluedye exclusion, acridine orange staining or MTT-turnover)
